Abstract:
RIO Kinase is the family of proteins being studied in Dr. Nicole LaRonde’s biochemistry research lab on the University of Maryland campus. These proteins are imperative for cell function; cells die without them. Rio proteins play an important role in processes in the cell such as ribosome biogenesis. In the research lab, these proteins are being studied in order to analyze their structure and function. Once their structure and function is known, drugs can be made to activate or repress the expression of these proteins. These drugs can hopefully stop protein caused diseases such as Huntington’s. The processes to determine the structure and function of these proteins are very common in the lab. This manual will provide procedures and background information on determining the structure and function of these proteins. The procedures include growing small-scale cell cultures to produce cells expressing the protein, making buffers for the protein purification process to isolate the desired protein, and running an SDS PAGE. The manual will provide all students working in the lab with a guide with these standard procedures. It will give the researchers a better understanding of what they are actually doing and why, and allow them to become more familiar with these procedures. In addition, new procedures can continue to be added to this manual over the years as the lab develops new procedures and processes.

I imagine a reader that is slightly uncomfortable and nervous by coming into this new lab. I imagine the reader being overwhelmed with all of these procedures, but appreciative for the manual being there.

Reader’s Response:
I don’t know where the materials are in the lab. I’ve already been told to perform one of these procedures but do not know where to start. Hopefully this works out. I think it should as long as I follow the procedure exactly. Is it bad if I still ask questions even if I had the procedure in front of me? But the graduate students will be very impressed if I successfully perform this procedure. Hopefully the person who wrote this was right..

D, can you include at FAQ section that address some of the reader's concerns: location of equipment, etc.?

You can address the other concern -- asking questions -- in your cover or closing note.
